the thermostable mutant TdT3-2 shows a 10 C increased melting temperature as compared to the wild type enzyme. The mutant is capable of incorporating higher amount of ddCTP onto double stranded blunt end DNA
L397S
natural variant of TdT, impaired polymerase activity in vitro and impaired N-additions in an in vivo V(D)J assay
T450S
natural variant of TdT. Mutation leads to a decrease in the amount of product
additional information
transgenic mice lacking either long or short isoform or both, short isoform catalyzes enzymic reaction, long isoform modulates its function

mutational analysis demonstrates that the N-terminal portion of Tft, including the BRCA-1 C-terminal domains, is nor required for TdT activity, although the BRCT domain clearly contributes quantitatively to N-nucleotide addition activity. The second helix-hairpin-helix domain of TdT, but not the first, is required for activity. Deletional analysis suggests that the entire C-terminal region of TdT is necessary for N-nucleotide addition in vivo
