GENERAL STABILITY
ORGANISM
UNIPROT
LITERATURE
activity of the recombinant enzyme (MtPGDH) is almost lost within 24 h after purification when it is stored in 20 mM phosphate buffer, but not in 100 mM phosphate buffer or in 20 mM phosphate buffer containing 100 mM sodium chloride or potassium chloride
at low ionic strength the enzyme irreversibly loses activity with time. In 20 mM phosphate buffer, pH 7.5, most of the activity is lost within 24 h. The activity loss is prevented if the ionic strength is kept above approximately 100 mM salt
-
dilution inactivates
dithiothreitol increases stability
enzyme is highly thermostable
-
inorganic ions, i.e. Cl-, SO42-, HPO42-, essential for stability
-
phosphate ions stabilize the enzyme
