Cloned (Comment) Organism
construction of a plasmid generated to facilitate double homologous recombination to remove the DNA encoding C-terminal transmembrane domain from the genomic locus of TgElp3, transient expression of recombinant YFP-tagged in RHDELTAhx parasites from a vector construct using the tubulin promoter and containing anHXGPRT selection marker Toxoplasma gondii

Localization

Localization Comment Organism GeneOntology No. Textmining
mitochondrial outer membrane due to possession of a unique C-terminal transmembrane domain, the enzyme localizes the protein to the mitochondrion. TgElp3 is devoid of known mitochondrial targeting signals. The enzyme is oriented with its catalytic components facing the cytosol and its C-terminal transmembrane domain inserted into the outer mitochondrial membrane, consistent with a tail-anchored membrane protein. The mechanism of trafficking to the outer mitochondrial membrane involves a unique C-terminal transmembrane domain present only on Elp3 homologues of apicomplexan protozoa and other select chromalveolates. TgElp3 is essential for parasite viability and must be positioned at the mitochondrial surface Toxoplasma gondii 5741

General Information

General Information Comment Organism
physiological function Elp3 is the catalytic subunit of the well-conserved transcription elongator complex. Apicomplexa lack all other elongator subunits, suggesting that the Elp3 in these organisms plays a role independent of transcription. Enzyme TgElp3 is essential in Toxoplasma and must be positioned at the mitochondrial surface for parasite viability Toxoplasma gondii
