gene PIM1, recombinant overexpression in enzyme-deficient trangenic Pim1-Tx Mus musculus driven by vav hematopoietic promoter/regulatory elements. Compared to wild-type littermates, Pim1-Tx mice show enhanced hematopoiesis as demonstrated by increased numbers of Lin-Sca-1+c-Kit+ (LSK) hematopoietic stem/progenitor cells and cobblestone area forming cells, higher BrdU incorporation in long-term hematopoietic stem cell population, and a better ability to reconstitute lethally irradiated mice. Pim1-Tx mice express human PIM1 mRNA in all hematopoietic tissues including spleen, bone marrow and thymus, but not in non-hematopoietic tissues such as kidney | Homo sapiens |

additional information | gene PIM1, recombinant overexpression in enzyme-deficient trangenic Pim1-Tx Mus musculus driven by vav hematopoietic promoter/regulatory elements. Construction of Pim1-/-, Pim2-/-, Pim3-/- single knockout mice. hematopoietic stem cells from Pim1-/- KO mice show impaired long-term hematopoietic repopulating capacity in secondary and competitive transplantations. The defects are not observed in hematopoietic stem cells from Pim2-/- or Pim3-/- knockout mice. Limiting dilution competitive transplantation assay estimated that the frequency of LSKCD34. hematopoietic stem cells is reduced by about 28-fold in Pim1-/- KO mice compared to wild-type littermates. Approximately 10% of Pim1-Tx mice develope acute lymphoblastic leukemia/lymphoma between 141 and 435 days of life while wild-type littermates are leukemia free in entire lifetime. Hematopoiesis-associated genes are affected by Pim1 kinase. Phenotypes, detailed overview | Mus musculus |
