Trichoderma reesei two truncated genes (Man1DELTACBM lacking carbohydrate-binding module domain and Man1DELTALCBM lacking linker and CBM domains) from Trichoderma reesei Rut C30 are cloned and expressed in Pichia pastoris. The expression efficiency is improved after truncating carbohydrate-binding module. Activities of Man1 and Man1DELTACBM in the culture supernatant after 168 h of induction are 34.5 and 42.9 U/ml. A value of 0.36 U/ml is detected for Man1DELTALCBM (lacking carbohydrate-binding module and linker). Man1 shows higher thermostability than Man1DELTACBM at low temperature, whereas Man1DELTACBM has a higher specificity for galactomannan (Km: 2.5 mg/ml) than Man1 (Km: 4.0 mg/mL) additional information
